I have slight Wi-Fi signal down at my barn from my home Wi-Fi. If I were to buy a signal booster and set it up in the barn, would I regain full signal at the barn from my home Wi-Fi router?

Yes, a good signal booster can help to improve the signal for smooth networking. Though a good environment of Wi-Fi system is a better way to use maximum resources.

A lot depends on the booster. Some have a better range than others. How far are we talking about from the barn to the current router?

Moreover, make sure the network (that you are using) is as fast & reliable as possible is to use upto date hardware specially if you are streaming so many HD video to devices like Chromecasts/Smartphones. In common practice, routers are often confined to particular locations based on where the internet connection enters your home & connects to the modem. Make sure you are using Quality of Service (QoS) to to reign in those bandwidth hogs.
