In my Microsoft Excel spreadsheet, I have three columns that I want locked (so no one can change anything). But when I clicked on format -> Lock Cells, it's protecting all of the columns. I just want these three locked. How can I do this?

Just click on the columns you want to protect. Right click and select format cells.
On the protection tab check the box LOCKED. Then ok. Next step is to click the REVIEW tab on the top and in the CHANGES section select PROTECT SHEET. In this pop-up window make sure the protect worksheet and contents box is checked. Set a password to unprotect the sheet and allow changes to be made. Then you can select other protection options. When done click OK and save your worksheet.

At very first, unlock/un-protect all of the cells on the sheet (if any). To do this, select all of the rows/columns in the sheet, then right click & select format cells from the popup menu. Here you can select the protection tab & deselect locked checkbox. Again, select cell(s) that you wish to lock/ protect. Bring back the pop up menu as described earlier. on format cells, select the protection tab. Select the checkbox of locked and click OK. To take effect, protecting worksheet is also necessary. Now select the review tab from the toolbar & click on protect sheet button. On next dialog box, select the checkbox(s) according to your wish. Press OK.

Here is a good link from Microsoft: https://support.microsoft.com/en-in/help/214081/xl-how-to-lock-individual-cells-in-a-worksheet

But as you did not reveal version, so it may slightly vary.
