More information is needed. Have you updated the system very recently? Is there any anti virus installed either external? Is there any pending update? Have you checked your User account? Is it Standard/ Administrator?

what files are you trying to access that are causing problems? What are you trying to do with these files open them, modify them , delete them or something else ?

Since Microsoft makes these decisions for Windows system files, it's likely that only Microsoft can provide a viable answer. Without knowing which files, first guess is that even Administrator can't access them while the system is running. Accessing them would interfere with the system operation.

There can be any number of files that shouldn't be touched unless the system is quiescent. Access may be possible if the hard drive is attached as a secondary to a different system.

It is also possible that that Microsoft or another program hid them during updates, and did not release them properly when the updating completed.  I would have agree with TheRealRaven on checking with Microsoft for the Answer.

If you have updated the operating system recently, might be this is your issue. Go to Update history & try to uninstall the recent updates be carefully. But before doing that, be cautious about your files/ data etc.

One more thought, you can try to access in Safe mode with Admin rights. Though, there are some restrictions in Safe mode, but at least you may try to access these files, which were not accessible in Normal/ General  mode.

It is also possible that they are now hidden.  When you are in file explorer is the check mark showing in the show hidden files.  If not place it in the check box.
