Getting an error message with ASP.NET MVC application in Windows Server 2008 R2

I've recently installed .NET 4 on Windows Server 2008 R2 x64 and I keep getting this 500 Internal Server Error with an ASP.NET MVC application. It was running great on 3.5. I have custom errors turned off in web.config but no detailed error is showing, just the internal error. Anybody have an idea why this is happening?

    In those cases it usually helps to run the application from the server, which helps to get a more detailed error message, and a stacktrace.

    Without looking at the detailed error, it would be really difficult to figure out what the issue might be.
