File system block size vs disk block size

Take that my file system block size is 4k, and my disk block size (physical sector size) is 4k. What happens if my increase file system block size to 64k?
Is file system block size actually I/O block size, or is it something else?
And what is kernel block size?
How does it all impact my reading and writing to, let's say, SATA disk on /dev/sda?
