What would be the fastest way to access files?

I have about 750,000,000 files (I know it's alot) that I need to have stored. The biggest thing for me is that I need to access these files randomly, at any given time in the shortest way possible. What should I do to accomplish this? One of my coworkers said to organize them into several directories. Would that be feasible? Thanks for the input.

    You must categorize them by directories! There are performance problems of many sorts with very large directories. How much depends on the OS and implementation details. Not to mention the fact that at some point a real person will need to find/verify/inspect some files.
