 One-to-One Back Up

I am currently using an IBM T42 with a 60GB Hitachi Drive. I recently purchased an Ultra-Bay Slim with a HDD adapter and bought the identical drive as well. I would like to be able to create a back up of my primary drive such that whenever it is down, I can plug in the secondary drive and be on the way. My confusion is that most back up programs create images of the primary drive. I assume that I will not be able to pull out and plug in the secondary drive and start straight away. Any advise? Also, which backup progam would suite my need. One more thing, is there a real diff between NTFS and FAT32?
