 How do I identify the protocol to select in HP LoadRunner?

I believe this is a very tool-specific question, but then it might be scalable to other tools, too. I have an application that contains different technologies such as a mainframe backend, IBM UDB DB, and a .Net front end. My question is: How do I identify the protocol to select in LoadRunner, or what are the questions that I need to ask for approaching the performing testing of this type of application. Thank you.
