We currently have a mail server (POP3) in-house (Rocliff-Mailsite); an email originated in our as400 is routed to this server which in turn will relay the msg to the end destination. We're moving to an MSExchange mail server
and I'm trying to implement the same approach with this new server the same way but it doesn't work. I have the IP address of the server but when I try to telnet via port 25 it just do not respond.
What should I be looking for to make this work?

I got this back from our exchange server provider:
‘your AS/400 server is not allowed because the process will require authenthication
and SSL. We do not support open smtp relay.’
What should I do now? Are there any settings on the AS/400 to provide the authentication and SSL they’re asking?
Several years ago we purchased MailTool from BVS Tools. It solved all our e-mail problems on the AS/400. IT works great. We use it a lot. One example is an e-mail message sent to my cell phone as the AS/400 nears the end of an IPL. Most of our IPLs run un-attended in the early morning hours. Seeing this confirming message let’s me know all is well.

The important part seems to be “…our exchange server provider.”
That is, it’s not “Exchange” that is at the center of the question. The issue is that your “server provider” is unwilling to accommodate your need. Thet would not have to create an ‘open relay’ that was totally open to the world. All they should have to do is accommodate just your AS/400 IP address. It seems that they’re not up to the (fairly basic) level of knowledge that allows that to be done. IMO, they should be able to assist in setting this up.
Switch providers.
