 Do I need to remove SQL Server 2005 to install SQL Server 2008 R2?
Do I need to do a complete removal of 2005, as tried that on a new Windows Web server 2008, as wanted to put 2008 SQL Server.

No - you can do an in place upgrade directly from SQL Server 2005 to SQL Server 2008 As with any database server upgrade you should test your application against the new database version before upgrading to ensure that the application works correctly with the new version. While it isn't likely that there will be a problem, it is possible.
