Ultimately you want to co-locate all of these servers together. CAS and UM need to be close to the MBX for access reasons and hub needs to submit to mailbox, so you should keep them together.

As long as they can access each other over the network without delay then it should not matter if they are in the same location or not. Depends on your needs and if a single administrator needs to have access all in one place or not.
