how do I set up an automatic reply to sender on Lotus Notes.

There are a variety of ways to do this. Perhaps the most basic is to create an agent and use the Simple Action called "Reply to sender."

You have to take into consideration when doing this not to cause a Mail Loop – This would cause a huge increase of mail if another agent is set up in the same way. OUT OF OFFICE agents have a check that states a check to make sure that it will not reply to the same address.

So a Simple agent might not be correct in the long run but it would work.
